uncode_shortcode_id=\u00a0\u00bb360120&Prime;][vc_custom_heading uncode_shortcode_id=\u00a0\u00bb154348&Prime;]Essais de performance en culture cellulaire[\/vc_custom_heading][\/vc_column][\/vc_row][vc_row][vc_column width=\u00a0\u00bb1\/1&Prime;][vc_column_text uncode_shortcode_id=\u00a0\u00bb166228&Prime;]<\/p>\n<p class=\"font-claude-response-body break-words whitespace-normal\">Afin de d\u00e9terminer si le reconditionnement influe sur les propri\u00e9t\u00e9s de culture cellulaire des plaques en polystyr\u00e8ne \u00e0 6 puits (cuture coated), nous avons \u00e9valu\u00e9 trois param\u00e8tres : la morphologie cellulaire, la confluence cellulaire et la viabilit\u00e9 cellulaire, en utilisant des fibroblastes Rat-1 comme mod\u00e8le.<\/p>\n<p class=\"font-claude-response-body break-words whitespace-normal\">Les plaques ont \u00e9t\u00e9 soumises \u00e0 un maximum de cinq cycles de reconditionnement. Apr\u00e8s chaque cycle, deux plaques ont \u00e9t\u00e9 mises de c\u00f4t\u00e9, et les plaques restantes ont \u00e9t\u00e9 r\u00e9ensemenc\u00e9es puis renvoy\u00e9es pour reconditionnement. Une fois tous les cycles termin\u00e9s, toutes les plaques (y compris deux plaques neuves servant de t\u00e9moins) ont \u00e9t\u00e9 ensemenc\u00e9es simultan\u00e9ment \u00e0 partir de la m\u00eame culture cellulaire afin de minimiser la variabilit\u00e9 et de permettre une comparaison directe.  <\/p>\n<p class=\"font-claude-response-body break-words whitespace-normal\">La confluence et la morphologie ont \u00e9t\u00e9 \u00e9valu\u00e9es par microscopie \u00e0 contraste de phase \u00e0 des grossissements de 4x et 40x. La confluence a \u00e9t\u00e9 quantifi\u00e9e \u00e0 partir d\u2019images \u00e0 4x \u00e0 l\u2019aide du logiciel ImageJ, tandis que la morphologie a \u00e9t\u00e9 analys\u00e9e \u00e0 partir d\u2019images anonymis\u00e9es \u00e0 40x en mesurant la surface cellulaire, le rapport largeur\/hauteur et le diam\u00e8tre de Feret sur 10 cellules s\u00e9lectionn\u00e9es au hasard par image. La viabilit\u00e9 cellulaire s\u2019\u00e9levait en moyenne \u00e0 93 % sur les plaques neuves et variait entre 81 % et 98 % au cours des cycles de reconditionnement. La confluence s\u2019\u00e9levait en moyenne \u00e0 87 % sur les plaques neuves et \u00e0 83\u201393 % sur les plaques reconditionn\u00e9es. La surface cellulaire individuelle s\u2019\u00e9levait en moyenne \u00e0 889 \u03bcm\u00b2 dans les plaques neuves et \u00e0 798\u2013898 \u03bcm\u00b2 dans les plaques reconditionn\u00e9es, tandis que les rapports largeur\/hauteur \u00e9taient pratiquement identiques entre les deux conditions. Les images microscopiques n\u2019ont r\u00e9v\u00e9l\u00e9 aucune diff\u00e9rence apparente dans la morphologie ou la r\u00e9partition des cellules entre les plaques neuves et celles reconditionn\u00e9es jusqu\u2019\u00e0 cinq fois. Aucune diff\u00e9rence statistiquement significative n\u2019a \u00e9t\u00e9 observ\u00e9e pour aucun de ces param\u00e8tres.     <\/p>\n<p class=\"font-claude-response-body break-words whitespace-normal\">La variabilit\u00e9 \u00e9tait similaire tant pour les plaques neuves que pour les plaques reconditionn\u00e9es, et n\u2019a pas remis en cause la conclusion g\u00e9n\u00e9rale : le processus de reconditionnement n\u2019a pas eu d\u2019impact significatif sur les performances de la surface des plaques. Notre proc\u00e9d\u00e9 permet donc d\u2019utiliser des consommables reconditionn\u00e9s jusqu\u2019\u00e0 cinq fois pour le passage et l\u2019ensemencement de cellules. Veuillez noter que cette conclusion s\u2019applique aux consommables ayant subi un traitement de surface chimique (type culture coated), et non \u00e0 ceux ceux ayant subi un traitement de surface biologique, tels que les consommables recouverts de collag\u00e8ne.  <\/p>\n<p class=\"font-claude-response-body break-words whitespace-normal\">Ces r\u00e9sultats concordent avec ceux de recherches ant\u00e9rieures. Trusler et al. ont montr\u00e9 que la r\u00e9utilisation des P\u00e9tri de culture au cours de plusieurs passages n&rsquo;avait aucune incidence sur la morphologie, la prolif\u00e9ration ou la viabilit\u00e9 des fibroblastes pulmonaires humains ( <em>Front. Sustain., <\/em>  2024). De m\u00eame, Clancy et al. ont rapport\u00e9 que la r\u00e9utilisation de P\u00e9tri de culture dans le cadre d&rsquo;exp\u00e9riences sur des embryons <em>de Xenopus laevis<\/em> donnait des taux de f\u00e9condation et de survie comparables \u00e0 ceux obtenus avec des P\u00e9tri neufs (<em>Differentiation,<\/em> 2023).
